As far as we know, it should work. But it has only been designed for 4TB hard drives (which doesn't necessarily mean it won't work). So we can't say 100% yes unfortunately.

Yes. Note from Snyology: This drive is only compatible with Synology NAS models with hard drive tray shipped since June 2014. Please check if your model was shipped after the above date.
I installed it in my old Qnap four-disk NAS with other machines running next to it and didn't notice any noise level difference.

Yes, of course :-)! Even if it would "run all the time" (i.e.: as intended) - what does it mean to "run all the time"? The disk goes into sleep mode anyway, so it's stationary. It would be stupid not to be allowed to switch it off ;o)!
ps: The 6TB would also do in terms of capacity...
They should run without problems on any Sata port.
