I have an iMac 21.5 from mid 2011, which seems to have a Thunderbolt port. Can I connect this drive to the first generation of Thunderbolt (I think Thunderbolt 2 or 3 is available now)?

Yes, you can. You can connect this drive directly to Thunderbolt 1 or Thunderbolt 2.
