Can I run the drive via the USB port of my Surface Pro 6 or is the voltage sufficient?

You need two USB 2 ports for this drive. If you only have one free, then you just have to take one of these or see if there are external drives with USB 3. The voltage for USB 2 is always 5 volts, for USB 3 up to 20 volts.
