Can this 12TB drive be used together with a Samsung 860 QVO (4000GB, 2.5") in a Mac Pro 5.1 (built 2010)? Or what is the maximum size of a backup HDD that can be fitted, in one of the four HDD bays?

If I see this correctly, standby and idle mode were confused here because idle mode = 5 watts and standby/idle mode is not specified in the data sheet, which makes no sense for a server (data centre) disk that is supposed to work.
