Are we supposed to remove the plastic plug that's at the bottom in the middle?

The plastic plug in the Kibernetik Party Cooler KS90M normally serves as a drain plug. This plug should be removed if you want to clean the cooler or drain water. However, if you are using the cooler in normal operation, the plug should remain in place to prevent liquids from leaking out.

Can I remove the basket (grille) from the fridge and place the bottles directly on the bottom?

The basket can certainly be removed. You can then place the bottles upright, which is not possible in the basket. I've never done this myself, but I don't see a problem as long as the air can circulate inside.
