Is it possible to extend the camera compartment from the 15cm depth to 16cm in a single place? have a slightly longer lens

I think you can. But since the opening to the main compartment is on the inside, i.e. at the back, you will feel that something doesn't quite fit. It's more likely to press on the back than on the other side. I would pack the lens lengthways.
